my inclination would be to start with; sound sources top left, followed by sound modifiers top right, control voltage sources below.
something like:
VCO-VCO-VCO-Noise-Mixer-VCF-VCA-Mixer-Output
MIDI-LFO-Blank-Blank-LFO-ADSR-ADSR
Are the connecting jacks on the bottom of each panel? If they are on top, I would reverse the rows.
It really depends on what your favorite patches are.

I am building a modular synth using MOTM sized modules. My cabinet is 20U wide and 10U high. I am wondering if there is a preferred layout of modules in a system that makes best use of the patch cables (keeping them short and as tidy as possible while maximising access to the front panels) as well as conforming with the more common ways the modules are used together?
For example, would you group all modules together, for example :-
a.. all the VCOs, then the VCA.s then the VCFs......
or would you group them for optimum operation like, for example:-
a.. VCO - [ADSR - VCF] - [ADSR - VCA]......., or
b.. VCO - VCO - VCO - MIXER - [ADSR - VCF] - [ADSR - VCA].........
My system currently consists of:-
1.. 3 x VCO
2.. 1 x VCF
3.. 1 x VCA
4.. 2 x ADSR
5.. 2 x VC-LFO
6.. 1 x NOISE
7.. 2 x 3-channel mixer
8.. 1 x MIDI-CV
9.. 1 x Output Module (tone controls and headphone amp)
