[sdiy] Knob Box Limitations?

I've been lurking here for a couple weeks hoping my question would come up,
yet it hasn't... I don't know a whole lot about Synth DIY, I've been
together a couple PAIA kits, but nothing really intensive, and nothing that
didn't hold my hand the entire way.
The project is this... a friend and I are in the process of building a
sequencer, the software part is nearing completion and now it's time to
start working on the hardware interface. Basically, it's just going to be a
huge knob box that'll send parameters to the computer, and then out to the
synths... The box is going to have several hundred knobs (at least 300) and
I'm wondering if I'm going to run into problems with MIDI... I'd prefer to
use something like ethernet/osc/usb/etc but I don't know how easy this is to
implement or if that would solve any problems.
I've looked at ucapps, but it doesn't say much about the maximum number of
knobs you can have... if a couple hundred isn't a limitation that looks like
a great existing format to work with.
