These are good questions but you don't need to ask someone else for the answers. Take 3 minutes and do a test and find out first hand. Open a color image, convert to mode->grayscale, and save it to a new filename. Then reopen the color image, go to channel mixer, monochrome + twiddle some settings, then convert to mode->grayscale (you'll see whether or not it changes your settings) and save it to another new filename.
