There is no one best answer as it depends on the neg, subject & scanner.
I always scan in RGB on my drum scanner but from there I will either pick the best channel deleting the others or simply convert to b&w in Adjustments.
You may find your own method that works best for your setup that differs from mine.

Move to quarantaine

This moves the raw source file on disk only. The archive index is not changed automatically, so you still need to run a manual refresh afterward.
