Calibration Mode with inkseparation6.tif

If I'm using a six colour printer (R300) should I be using the inkseparation6.tif file instead of the default? Could someone explain how the overall calibration is different for a six ink printer compared to the one outlined in the tutorial for an eight ink printer? Can I just use Black, Yellow, Light Cyan, Light Magenta to produce grey tones? Anyone using the Epson R series please feel free to add comments.
