I believe the color contrast filter and the film type sliders of Convert BW Pro require color layers in order to be effective - once all three layers are equal in density (which I believe is the case once the image is in grayscale) there is nothing for the color contrast filter or "film type" selector to differentiate between the layers. The exposure and contrast sliders probably would still work, but to preserve the full utility of Convert BW Pro you probably would need to stick with your option 1 rather than use option 3.
