I recently sample a section of a song I really like using the EXS24.
Now I want to create a beat around the sample. I was able to paly the sample via keybord 
and record it on the arrange window. However every time I play a beat around the sample 
it's off! Please advise

Why don't you just import the song into the audio window then onto the arrange page. Then find a section with 1 or 2 or 4 bars of the beat you want. Set the locators to loop. Open the sample editor and adjust the start to be right at the start of a beat and the end of the loop to go right up to the start of the next beat (just before it). Then cut the audio according to the locators so you have the proper looping sections.

Next count the bars. (E.g. 12 beats is 3 bars). Then highlight 3 bars on the locator bar - regardless of whether it agrees with the section or not. Now select the cut piece of loop, and use the key command to set the tempo globally by object length. This will set the tempo properly.

Next you can either manually isolate the beats, or use the audio to midi function to create a midi region that mimics the beats. Then move that midi file to a track with a drum instrument (Ultrabeat, Battery, RMIV, etc). Next mute the original file and see how it sounds.

You can then use that midi as a template for a groove that you can quantize all other midi too.

Move to quarantaine

This moves the raw source file on disk only. The archive index is not changed automatically, so you still need to run a manual refresh afterward.
