Well, typically on calibrates the monitor first, since its your eyes in the printer profiling process...

That should be reasonable for a moderately lit studio area...

The Ambient Light Tool would adjust your monitor "brightness" to your ambient light. The Measured mode would allow you to enter a different white luminance (AKA: Brightness) value to match your proofing light, if desired.

5. What do I do if I need to tweak the moniter profile to get it to
better match the print I've made with my custom profiles?


Other way around: unless you have chosen inappropriate target values for your display profile, you should tweak the softproof of your printer profile B&W values (in the Ref Black and Ref White options in Spyder3Print/PFP2) or the print output of your printer profile by using the basic or advanced sliders.


Do I
adjust gamma and white point settings in the Spyder software and
recalibrate?


Adjusting gamma won't change much, as applications compensate for gamma, so it will look the same. Adjusting whitepoint can change your percieved white tone on screen; that can be good, depending on what proofing light brightness, or what type of paper you are using (whitened paper has a cooler whitepoint).

6.Could you please be more specific as how to adjust the white
luminance?

Depends on your goal. First read all the help on each related screen in your software. That covers a lot of this, in a lot of detail.
