I'm using the first spyder and try to calibrate a two monitor system
hooked onto the same video card, one monitaor at the digital connector
the other at the analogue adapter. My primary monitor does not load
the profile instantly. I have to reboot the PC. Is there a problem
using the old spyder with a digital monitor interface? Any suggestions
how I could activate a new profile and compare it to another one
instantly without rebooting?

This is a recipe for disaster in more than one way. The OptiCAL and PhotoCAL software from the "first Spyder" is pretty ancient these days, and there may well be issues with loading video corrections, etc. in current OSes. Also, assuming that you get get distinct video corrections for two displays from one card's analog and digital outputs is pretty questionable. So I would expect you can color manage one of your two screens, but not both.

You are mistaking loading the video corrections for loading the profile; the profile is only loaded by color managed applications like Photoshop; what you see at start up is the video corrections being loaded to the videocard by the ColorVision startup utility (since Windows does not automatically do this for you). There may be only one slot, for one set of videocorrections, in your card.

Not universally, but I would expect issues to be sneaking in over time...

You have some incorrect assumptions here. The OS does not use the profile, only color managed apps do that. What you wish to compare could be the video corrections, the profiles, or (ideally!) both, since mixing them up will give you incorrect results on screen. I don't believe that flexible comparison of profiles one to another will be possible with the products you have.
