Just be sure to backup your original file. If I were going to do it the way you're trying to, here's how I'd do it: First I'd look at those beat slices and note at which sample each one starts. Then I'd create a new instrument. Then I'd create however many zones I need, probably mapped only to white keys. Then I'd load the sample to each zone and disable pitch (just to be safe). Finally, I'd set the start point of each zone at the sample the beat starts and end point at the last sample before the next beat. Of course, you'll want to alter attack/decay time from the front of EXS so you avoid "surprises." I've pondered making construction kits, but one life is just too short to get to everything. The problem with the above method is that the same long sample would be loaded multiple times, decreasing your sample RAM. You might as well chop it up. I'm not sure modulation source/destination will work, although technically it SHOULD, because there's the question of accurately starting the sample at the correct point. If you have, say, 4 beats and the keys were set to trigger a percentage modulation, like 0%, 25%, 50%, and 75% respectively, and each beat occurs at EXACTLY those points, then there shouldn't be any problem. Just set it up in the EXS router. Or you could just keep things simple and chop it up. ;) I've never tried it your way before, but I think I'll check it out to see if it CAN be done.
