If it is the first time you are loading the EXS24 instrument the long delay
is because Logic is searching for the sample files on your hard-drives. You
can speed this up on a system with multiple drives by specifying the drives
to be searched in the EXS preference  (Edit/Preferences when the EXS
instrument editor is open)

On subsequent loads the loading time should be quite fast if your drives
are fast -- ideally they would be at least 7200 rpm and set to DMA in
Device Manager -- look under properties for the hard-drive where your audio
file are located.

This moves the raw source file on disk only. The archive index is not changed automatically, so you still need to run a manual refresh afterward.
