I haven't used Lightroom before but I just downloaded it to try.
Unfortunately I've also run into quite a few weird occurances.

Since Lightroom does not appear to support grayscale, you are probably right
to try the RGB version of the QTR profiles. I received both the "invalid"
error and
also quite a few complete crashes of the whole program. I also decided to
see
what would happen with an ordinary Epson RGB profile - not that the results
would
be any good. But it crashed as well. So I suspect it's because of some
grayscale
issue rather than the profile. The crashes are all in the OS X Print Manager
so
help from Apple and/or Adobe will probably be needed.

I've played with various combinations and I think I've found a workaround
for
custom made QTR ICC profiles. It doesn't work with the generic QTR-RGB Matte
Paper.

Here's what I did:
1) Make your custom ICC using the RGB version as before.
2) In Lightroom, select the Epson driver as the printer. Setup the ICC
profile in
the Color Management section. Hit Print.
3) Instead of printing in the Print Dialog, hit Preview. This will output a
pdf file to the
Preview program.
4) Now you can print from Preview, select the Quad printer and all the
options you
want and print. 

The idea is Lightroom applies the RGB profile as normally, and Preview
handles the
conversion to grayscale.
