My guess from this is that you are on Windows, and have one of those problem systems where calling Windows to set the profile as the OS monitor profile fails... (and Windows also fails to send back a notification when this happens...). If so, then use the Profile Chooser software that comes with Spyder2PRO to assign the profile as the monitor profile.
