The samples for you libraries can reside anywhere, as can the sampler instruments as long as you create an alias to their location from the default "Sampler Instruments" folder. Once everything has been moved, EXS will have to search for the samples again the first time you load each instrument, which can take a long time, but will speed up after the initial find. Project Manager can re-link everything, but I understand this process can take thousands of years. EXS Manager, on the other hand, can handle this task even for gargantuan libraries in just minutes---or less. If you've got a "hefty" library as you mention, you really should have EXS Manager.
