The click is probably the result of a bad loop point. It's probably a small audio sample thats got a glitchy loop point. The "click" could be coming form either the start of the sample or the end (ie..as it loops to beginning..it clicks, but the click could be the result of an issue at the end of the sample "or" the beginning (starting point). It always sounds like it's the start of the note because it occurs at the "loop". If you hear the click as soon as you depress the key to play it, then it's probably a bad start point. If it starts ok..but quickly starts clicking over and over as you hold the note..then it's a bad loop point at the end of the sample. I'm not all that familiar with EXS editing, but hopefully, you can open the instrtument in EXS editor and futz with the loop points until you get rid of it. If not, try opening the audio sample file in an audio editor and looping it to check it for clicks. If there are no clicks..then the click is caused by bad loop points set in the EXS instrument. If you find clicks in the audio sample, take them out in the audio editor. Then, resave the file.
