- Open the EXS. on the top left there is a button called edit.
- Click edit
- In the window that opens up choose File -> New instrument
- Choose Zone -> Load multiple samples
- Browse to your audio file and click OK
- in the next dialog choose auto map (if it's only one file, read the 
doc for the other two options!)
- go File -> Save

Now you can load it up in the EXS pull down menu where your other 
instruments are.
