There's no way to do this as it stands.
The events that send a CC from the knob are handled by the sequence engine,
so if it's not running, they don't happen.
I have considered making the knobs in play mode programmable.
They can be enabled for sending 'mixer' messages (volume and pan
controllers) already.
Potentially you could set up the front panel as a proper control surface
which would work independently of sequence playback.
The problem with that is a lack of both code-space and RAM to hold much more
than one configuration of the 32 knobs.
You'd probably want at least one knob setup per bank.
