"Samples" are individual wave files (e.g. a C3-pp.aif sample from a piano), while "Instruments" are files that organise a collection of samples into a EXS-playable setup. Instruments are loaded from the black 'button' between the Options and Edit button in the main EXS interface. Samples are loaded in Zones in the Instrument Editor (which appears after you've clicked the Edit button).
