In the EXS24 instrument file, you can easily locate the sample path
but there are other important sample parameters that are also stored
in the instrument file, like the root key, the sample size, le loop point,
the number of channels, the resolution, and an offset to the beginning
of the audio data in a sample file. This means that you cannot just
replace one sample by another one like you did.
If you do that, then the EXS24 will aply the old parameters to the
new sample which may give inpredictible results...

Unfortunately a tabular version of the instrument definition -- something like
the one in Reaktor --  with "find and replace" type features has not been
offered. Perhaps a third party could offer this if Emagic will not. 

Often when building instruments from a set of similar sample sets,  the pattern
of naming files is followed over a series -- eg a series of drumkits with
identical layouts and file naming schemes. A routine which allowed the
substitution of a few characters would allow you to build subsequent
instruments in the series in seconds once the initial instrument was defined. 

Setting up graphical velocity switching layers would be pretty handy too.

Perhaps one of the enterprising utilities builders would be prepared to tackle
this project.
